The objective of this study was to characterize the clonal profile, virulence factors and antimicrobial resistance, particularly oxacillin resistance, of Staphylococcus aureus isolated from sheep milk. Milk samples were collected from all teats for the California Mastitis Test (CMT), somatic cell count, identification of S. aureus, investigation in these strains of genes encoding toxins (sea, seb, sec, sed, tst), biofilm (icaA, icaC, icaD, bap), leukocidin (luk-PV) oxacillin resistance by mecA gene detection and susceptibility testing (12 antibiotics). The aim of this study was to determine the prevalence of Staphylococcus aureus and risk factors for the acquisition of MRSA (Methicillin Resistant Staphylococcus aureus) as the main cause of skin and soft tissue infections. S. aureus were characterized for the presence of PVL, TSST-1 and mecA genes. SCCmec typing was carried out in mecA positive strains and PFGE was performed only in these strains. During the study period, 127 outpatients attending a dermatology clinical the Botucatu Medical School, a regional tertiary hospital in Botucatu, Sao Paulo, Brazil, were diagnosed with active skin infections. Biofilm formation is considered to be a selective advantage for Staphylococcus aureus mastitis isolates by facilitating bacterial persistence in the udder. It requires attachment to mammary epithelium, proliferation and accumulation of cells in multilayers. Abstract Staphylococcus aureus is an important cause of bloodstream infections. Therefore, the main purpose of this work was to characterize a collection of 139 S. aureus isolates from bloodstream infections in two public hospitals in relation to their antimicrobial susceptibility profile, staphylococcal cassette chromosome mec types, and clonal relationship. Determinou-se a concentração mínima inibitória (CMI) de ampicilina, cefalotina, eritromicina, gentamicina, neomicina, norfloxacina, oxacilina, penicilina G, tetraciclina e tilosina para 112 amostras de Staphylococcus aureus isoladas de infecções intramamárias bovina, em 33 rebanhos leiteiros da Zona da Mata do Estado de Minas Gerais. Foram isoladas 24 amostras de infecção clínica, 66 de subclínica e 22 de infecção crônica. As amostras de infecção crônica foram isoladas repetidas vezes dos mesmos quartos mamários de nove vacas de um rebanho, no período de 13 meses. The objective of the work was to study the microbiota of the conjunctival secretion of health professionals. Samples were collected from the clinically normal eyes of 40 health professionals in four different sectors, Proctology, General Intensive Care Unit (ICU), Male Ward, and Oncology of Hospital das Clínicas Dr. Augusto Leite, Aracaju, SE, Brazil. Ten professionals from each sector were selected. Amostras de músculo de carpa comum (Cyprinus carpio) e de água de três viveiros fertilizados com dejetos de suínos e de um viveiro em que os peixes foram alimentados com ração comercial para peixes foram analisadas durante seis meses. O objetivo foi observar a presença de bactérias patogênicas, como Salmonella, Staphylococcus aureus e coliformes totais e fecais, em peixes comercializados in natura no município de Chapecó, Santa Catarina. Este trabalho teve por objetivo caracterizar a produção da toxina da síndrome do choque tóxico (TSST-1) e de enterotoxinas estafilocócicas (SE) A, B, C e D em 127 amostras de S. aureus, isoladas de amostras de leite proveniente de vacas com mastite no Estado de Minas Gerais, entre 1994 e 1997. A verificação da produção de toxinas foi feita pela técnica de sensibilidade ótima em placa. Das 127 amostras testadas, 60 (47%) eram produtoras de TSST-1 e 54 (43%) produtoras de SE, 38 amostras produziram SED (30%), 24 SEB (19%), 8 SEC (6%) e 4 SEA (3%). Estes resultados trazem preocupações quanto à saúde pública pela alta prevalência de amostras de S. aureus produtoras de TSST-1 e de enterotoxinas em isolamentos a partir de leite de vacas com mastite. |

Realizou-se a detecção do gene de Staphylococcus aureus, de enterotoxinas e de resistência à meticilina com extração de DNA feita diretamente de amostras de leite. Das 200 amostras estudadas, 145 (72,5%) amplificaram o gene femA, e estas foram analisadas quanto à presença dos genes sea, seb, sec e mecA. Os genes das enterotoxinas mais prevalentes foram: sea (60%), seb (37,9%) e sec (6,9%). Foram encontradas 18 amostras de leite (11,0 %) com S. aureus portadores do gene mecA. A detecção de S. aureus diretamente do leite, sem a necessidade de isolamento bacteriano e a caracterização do potencial enterotoxigênico, demonstra que a técnica de PCR é muito útil para estudos epidemiológicos das infecções estafilocócicas da glândula mamária. This study aimed to detect methicillin resistant and slime producing Staphylococcus aureus in cases of bovine mastitis. A triplex PCR was optimized targetting 16S rRNA, nuc and mecA genes for detection of Staphylococcus species, S. aureus and methicillin resistance, respectively. Furthermore, for detection of slime producing strains, a PCR assay targetting icaA and icaD genes was performed.
